چکیده

The capacity of pituitary glands to release luteinizing hormone (LH) in response to LHreleasing hormone (LHRH) in vivo may be increased by previous frequent exposures to small quantities of LHRH. This phenomenon, called LHRH self-priming, may play an important role in the control of LH secretion. Our initial objective was to determine whether LHRH selfpriming occurs via a direct effect on bovine pituitary cells. Subsequently, we studied the effects of interval between LHRH exposures and source of pituitary glands (steers versus cows) on LHRH self-priming. Cells from bovine anterior pituitary glands were dispersed and mixed with Sephadex-G25, and the mixture was poured into glass columnS. Columns were superfused and effluent medium was assayed for LH. Challenges were .1 ng LHRH/ml or 0 ng LHRH/ml (sham) each for 10 minutes. In one experiment, quantity of LH released by a second exposure to LHRH averaged 124 + 6 ng/ml, which was greater than that released in response to an LHRH challenge (90 +4) given 40 min earlier. In a second experiment, variables were LHRH sequence (sham-LHRH versus LHRH-LHRH) and interval between challenges (40, 80, 160 and 320 min), Quantities of LH released in response to LHRH following the sham challenge or the first of two LHRH challenges were not different. However, LH released by a second LHRH challenge was 48, 39 and 48% greater (P<.01) than that released by the first when the interval between challenges was 40, 80 and 160 min, respectively. But when the interval was 320 min, no LHRH self-priming

Nitric oxide synthase in the rat anterior pituitary gland and the role of nitric oxide in regulation of luteinizing hormone secretion.

By using immunohistochemistry and in situ hybridization, we have demonstrated that the nitric oxide (NO)-synthesizing enzyme NO synthase is present in gonadotrophs and in folliculo-stellate cells of the anterior pituitary gland of male and female rats. A marked increase in levels of NO synthase protein and mRNA was observed after gonadectomy. In vitro studies on dispersed anterior pituitary cel...

P-122: The Effect of Beta Globin Intron on Human LH Hormone Expression in CHO Cells

Background Luteinizing hormone (LH) is a heterodimeric glycoprotein composed of alpha and beta subunits. This hormone is secreted from the pituitary gland. LH, in women triggers Menstrual cycle and ovulation. In men, LH stimulates production of testosterone, which plays a specialized role in sperm production. Up to day, LH hormone have produced in different ways such as codon optimization and al...

Cytoplasmic kinases downstream of GPR30 suppress gonadotropin-releasing hormone (GnRH)-induced luteinizing hormone secretion from bovine anterior pituitary cells.

GPR30 is known as a membrane receptor for picomolar concentrations of estradiol. The GPR30-specific agonist G1 causes a rapid, non-genomic suppression of gonadotropin-releasing hormone (GnRH)-induced luteinizing hormone (LH) secretion from bovine anterior pituitary (AP) cells. A few studies have recently clarified that protein kinase A (PKA) and phosphorylated extracellular signal-regulated kin...

Topographical localization of the receptors for luteinizing hormone- releasing hormone on the surface of dissociated pituitary cells

A derivative of the hypothalamic peptide luteinizing hormone-releasing hormone (LHRH) has been coupled to ferritin and the conjugate purified by gel chromatography. In its ability to stimulate the secretion of luteinizing hormone from pituitary cells in vitro, the conjugate has the same potency and specificity as the native peptide. When dissociated pituitary cells maintained in short-term cult...

Effects of STX, a Novel Estrogen Membrane Receptor Agonist, on GnRH-Induced Luteinizing Hormone Secretion from Cultured Bovine Anterior Pituitary Cells

STX is an agonist for a recently characterized membrane estrogen receptor whose structure has not been identified. We evaluated whether STX suppresses gonadotropin-releasing hormone (GnRH)-induced luteinizing hormone (LH) release from bovine anterior pituitary (AP) cells. We cultured AP cells (n=12) for 3 days in steroid-free conditions, followed by increasing concentrations (0.001, 0.01, 0.1, ...
